Don't know Edinburgh too well but I do know London. How long does your wife have in London and what kind of stuff does she like to do?

Waterproofs.

It's also the law in Scotland that you must wear tartan trousers if you are from America too, but I think Southern Americans are exempt from that.

Seriously, I'd recommend some warmish clothes for edinburgh, probably be OK in London though.

There's lots to see and do in Edinburgh, got an art gallery with loads of old spanish paintings on at the moment, so she might like that.

I'd recommend checking out some of the ghost tours, you can find the people who run them on the royal mile, they are quite interesting and they take you into old parts of the town that are supposed to be some of the most haunted places in Europe.

1 day is not really much time to see everything, so that and probably the royal mile and the castle would be what I'd suggest. If she is not too scared of heights she could maybe take a walk up the Scott Monument on Princes street, you get a really great view of the city from there.

There is a tour called The Duck which is an amphibious wartime vehicle which has been converted into a tourist bus. (This isn't a joke!)You pick it up next to the London Eye (which is worth going on itself) and it drives around the main central London landmarks, then it goes down a ramp into the Thames and takes you up and down, past the Tower of London and the Houses of Parliament.
Here's the links:

If she likes the Arts, the Tate Modern is an absolute must. The Victoria & Albert Museum and British Museum are fantastic too.
And a tip for a top class breakfast or lunch but not stupidly expensive is the Wolseley on Piccadilly - http://www.thewolseley.com

Oh, and the weather. She may be lucky and get some sunshine in London in September. But it could be cold and it may well rain. It may snow. Or there may be an unexpected heatwave. I suggest a range of clothing from short skirts to skiwear.
